LL-37 secretion system
LL-37, also known as hCAP18, is the only cathelicidin-like antimicrobial peptide found in human body. LL-37 is derived from the C-terminal region of the human cationic antimicrobial protein (hCAP), consisting of 37 amino acids at the N-terminal of Cathelicidin protein. LL-37 exhibits a variety of immunomodulatory functions. The BNUZH-China iGEM team 2022 designed a fusion protein consisting of IL-2 signal sequence (BBa_K4211002) and LL-37(BBa_K245114) for the secretion of LL-37 out of the cell. For protein purification and Western blot, we inserted a 6×His tag at the N-terminal of LL-37 gene.
Figure 1: Construct design: The LL-37 gene. The LL-37 gene was inserted after the CMV Promoter, and the secrete peptide gene IL-2 Signal sequence (BBa_K4211002) was inserted at the N-terminal of the LL-37 gene.
Usage and Biology
We used LL-37 to fight bacteria and prevent wound infection.
